Synthesis
The DNA Analysis Facility offers the synthesis of oligonucleotides through Sigma-Genosys, allowing you to enjoy the convenience of ordering through the DNA Analysis Facility coupled with the low pricing of large manufacturing and free shipping. The JHU Finch server allows you to easily keep a record of your orders, making it easy to reorder or to find concentration information. We offer four different synthesis scales and a variety of modifications.
All oligos are delivered dry and need to be resuspended. Oligonucleotides may be resuspended in either sterile water or, preferably, TE (10mM Tris, pH 7.5-8.0, 1 mM EDTA). The use of DEPC-treated water is not recommended. If you are making a concentrated stock solution we recommend dissolving your oligo in TE. Working solutions can be diluted from the stock solution using sterile, nuclease-free water.
0.025 µM
This is the smallest scale of synthesis, designed for oligos of 12-35 bases. It has a guaranteed yield of 3 O.D.s. It cannot be purified, but is desalted and quality control is preformed using MALDI-TOF mass spectrometry. This scale is shipped within 24 hours of ordering, which means that it will be returned to you within 48 hours.
0.05 µM
This scale yields a minimum of 5 O.D./150 ug when unpurified. It may be Cartridge, HPLC or PAGE purified for an additional cost. All purification methods reduce the amount of oligo returned. Purification method must be selected during the ordering process. This scale will take 48-72 hours to arrive at JHU. Purification adds 2-4 days to delivery time, depending on method selected. Oligos longer than 35 bases also may take up to 5 days to deliver.
0.2 µM
This scale yields a minimum of 12 O.D./ 360 ug when unpurified. It may be Cartridge, HPLC or PAGE purified. All purification methods reduces the amount of oligo returned. Purification method must be selected during the ordering process. This scale will take 48-72 hours to arrive at JHU. Purification adds 2-4 days to delivery time, depending on method selected. Oligos longer than 35 bases also may take up to 5 days to deliver.
1 µM
This is the largest scale offered through the DAF. This scale yields a minimum of 40 O.D./1,200 ug when unpurified. It may be Cartridge, HPLC or PAGE purified. All purification methods reduces the amount of oligo returned. Purification method must be selected during the ordering process. This scale will take 48-72 hours to arrive at JHU. Purification adds 2-4 days to delivery time, depending on method selected. Oligos longer than 35 bases also may take up to 5 days to deliver.
