Specialty Probes and Primers

Through our partnership with Sigma/Genosys, we are able to offer a wide variety of speciality synthesis products, including DNA probes (single and dual labeled), LNA probes, RNA oligos, custom siRNA, WellRED oligos, Molecular Beacons, LightCycler probes, and Scorpion probes, in addition to the "traditional" modifications such as inosine, biotin, phosphorothioate backbone, etc. All fluorescently labeled oligos are shipped in amber tubes to protect dye integrity.

Dual Labeled Probes:

Dual Labeled probes are linear probes that incorporate a 5'-end reporter dye and a 3'-end quencher dye. They can be used in many different applications, including real-time qPCR.

  • Pricing is a flat fee dependent on the labels chosen and includes HPLC purification.
  • LNA (Locked Nucleic Acids) can be added to your sequence to increase sensitivity and specificity.
  • Probes may be ordered in quantities of 1, 3, 5 or 10 ODs.
  • Label combinations are as follows:
    • 5' end 6-FAM, HEX or TET with 3' end TAMARA or BHQ-1
    • 5' end Cy3, Cy5, Cy5.5 or TAMARA with 3' end BQH-2
    • 5' end JOE, ROX or Texas Red with TAMARA or BHQ-1 or BHQ-2
  • Locked Nucleic Acids may only be used with a 5' FAM, HEX or TET and 3' TAMARA.
    • To add LNA bases to your sequence use the following codes: [+A], [+C], [+G], [+T]
    • A maximum of 8 LNA bases may be added per oligo.

Molecular Beacons:

Because Sigma-Genosys is a licensed supplier of Molecular Beacons (PHRI, NYC), these oligos can be ordered through the DAF. Molecular Beacons are probes that contain a 5'-end reporter dye and a 3'-end quencher dye designed in a stem-loop formation. These oligos are suitable for multiplexing, SNP analysis and allelic discrimination. They are typically used in real-time qPCR applications and are highly discriminatory.

  • Pricing is a flat fee dependent on the labels chosen and includes HPLC purification.
  • LNA (Locked Nucleic Acids) can be added to your sequence to increase sensitivity and specificity.
  • Probes may be ordered in quantities of 1, 3, 5 or 10 ODs.

    • 5' end Fluorescien, FAM, HEX, or TET with 3' end BHQ-1 or DABCYL
    • 5' end Cy3, Cy5, or Cy5.5 with 3' end BHQ-2 or DABCYL
    • 5' end JOE, ROX, or Texas Red with 3' end BHQ-1, BHQ-2 or DABCYL

LightCycler Probes:

ightCycler probes are optimized for use with the Roche LightCycler instrument. They are highly sensitive and sequence specific fluorescent real time qPCR probes. Several fluorophores are available for multiplex analysis. They are sold by Sigma under license from Roche Molecular Diagnostics, Inc.

  • Oligos are sold in pairs. Both oligos must be ordered at the same synthesis scale.
    • The first probe must have a 3' Fluorescein label.
    • The second probe may have the following modifications:
      • 5' end: LightCycler Red 610, 640, 670, 705
      • 3' Phosphorylation may be added to this
      • Internal: LightCycler Red 610 or 640 (not to be combined with 5' or 3' modification)
  • Locked Nucleic Acids may only be used with the Fluorescein probe, or with a 5' Red 640 and 3' Phosphate probe
    • To add LNA bases to your sequence use the following codes: [+A], [+C], [+G], [+T]
    • A maximum of 6 LNA bases may be added per oligo.

Scorpion Bi-Probes

Scorpion Bi-Probes is a duplex of two, complementary-labeled oligos, offering stronger signal intensity than the uni-probe structure. One oligo of the bi-probe is labeled with a 5' end reporter dye and carries the blocker and PCR primer elements. The other oligo is labeled with a 3' end quencher dye.

  • Available in lengths up to 45-mers

WellRED Oligos

WellRED oligos use cyanine-based fluorescent dyes with high extinction coefficients that absorb in the near-infrared region, thereby reducing background noise from biological materials, resulting in highly sensitive detection. WellRED oligos are sold by Sigma under license by Beckman Coulter, Inc. to be used CEQ Genetic Analysis Systems.
